Is Late-Onset Alzheimer Disease Spelled "ATV(N)"?

There is consensus that Alzheimer disease (AD) and AD-related dementias (ADRDs) are characterized primarily by proteinopathies of β-amyloid (A) and hyperphosphorylated tau (T) accumulation posited to underlie neurodegeneration (N). These hallmarks form the basis of the ATN framework for studying AD1 but are often insufficient to describe the composition of cerebral pathology present for the vast majority of older adults. The vascular contributions to AD, especially those that involve systemic and cerebral circulation, are important factors in our understanding of ADRD pathogenesis, yet they are often understudied.2
